Его творчество оказало существенное влияние на развитие поп-музыки второй половины XX века, блюза в особенности. Многократный лауреат премий «Грэмми».

Песни Джона Ли Хукера игрались во многих фильмах и компьютерных играх. Одна из компьютерных игр — «Mafia 2» — использовала его хит «Boom Boom». В игре «GTA vice city» по Хукеру был сделан один вид прохожих. Его лучшими друзьями были Карлос Сантана, Бонни Райт, Ван Моррисон, группа «Los lobos».
